In 1714, he became the pastor of the Woodbridge Church (First Presbyterian Church of Woodbridge). He was 28 years old at the time. His salary was 80 pounds a year. He continued to serve as minister there until 1752. He was one of the first members of the Board of Trustees of the College of New Jersey, which subsequently became Princeton University. He went to Mendham, NJ in 1753, where he stayed for the next 10 years as a minister. He remarried and lived on his wife's farm on Long Island, but returned to New Jersey upon her death and spent the last years of his life with his daughter, Elizabeth Greene, in Hanover.

Pierson Millenium listed that John died at the home of his daughter Elizabeth, married to Jacob Greene, in Hanover, New Jersey after a ministry of 56 years. He spent his early life in Newark under the instruction of his father and moved to Connecticut when his family moved back. He graduated from Yale in 1711, and went to Woodbridge, New Jersey to preach in 1714. He continued to preach there until 1752. He was on the first board of trustees of the College of New Jersey. He married Ruth Woodbridge. After his wife's death in 1732, he moved from Woodbridge to Mendham, New Jersey in 1753 where preached another 10 years. He married again to J. Smith and moved to her farm on Long Island, New York. After her death, he returned to Hanover, New Jersey to live with his daughter Elizabeth.
Abraham Pierson Descendants lists John marrying Ruth Woodbridge on 18 Jul 1716 in Piscataway, Middlesex, New Jersey. This source also lists John having children with Judith Smith (Hannah and David), but other sources do not list him having children in this marrige.

His history
John Pierson early days were spent in Newark New Jersey under the instruction of his father. He moved with his father to Conn. and graduated from Yale College in 1711. In 1714 he went to Woodbridge NJ to preach where he was ordained a Presbyterian minister April 29 1717, with a salary of 80$ a yr and the use of the parsonage. He continued in charge there until 1752 when he was dismissed at this own request in consequence of sme difference arising between him and his people. He was however dismissed without the least censure or imputation upon his character or conduct. He was justly deemed and reputed a worthy and able minister of the New Testament sound of faith of exemplary conversation and candor he well supported the dignity of his office and had but few equals in his day in theological knowledge. It was in his time and by his influence that the congregation obtained a royal charter and was incorporated under the name of the First Presbyterian Church of Woodbridge and did by their trustees take legal possession of a donation i land made them by the proprietors. He was one of the first board of trustees of the College of New Jersey. He removed from Woodbridge to Mendham NJ in 1753 where he remained for 10 yrs. and preached the gospel. He married again to J. Smith and moved to her farm on Long Island. After her death he returned to NJ. He spent his declining days at Hanover JN with his daughter Elizabeth wife of Rev Jacob Greene mother of Dr Ashbel Greene the 8th president of Princeton College where he died after a ministry of 56 yrs. Mr. Pierson Published a treatise on the Intercession of Christ and a sermon preached before the Presbytery of NY May 8 1751 on Christ the Son of God as God Man Mediator.
